Please help me in this if anybody can I will really appreciate.

I have been feverishly trying to sequence the BCR-ABL kinase domain using semi-nested PCR approach. Have tried following methods previously described in studies published by the Hughes group in Aus and various others but have been unsuccessful thus far! :-( I use the big dye sequencing V3.1 kit and ABI377…. any suggestions to help brighten ma day??? Have tried various concentrations of primers and template all of which yield a very noisy background but no signal!!! :-(

Can anybody help me out in this I will be really grateful.

I am a die hard researcher in leukemia since last 1 year, though I am a clinician in practice. My guide does work on CML and he does abl-kinase domain mutations and we have got good results. I can ask him to help you out, but before that I would like to know what exactly is your aim and how deeply you are involved in your study. Is it kind of working you doing just because it will get you a degree or you are really interested in such things? Can I get to know what kind of samples you are dealing with? Are they cases directly put on IM or after treatment with some other drugs? Currently there is a debate going on as to whether these mutations pre-exist before starting IM or other TKIs or they are a result of such treatment. Again whether this is because such mutated strains are selected over non mutant ones or they just develop by the drugs themselves!

First of all, thanks a lot for replying to my post! The aim of my research is for the detection of mutations in the bcr-abl kinase domain but I have been unsuccessful with respect to sequencing the domain so far. This is my research project for my post grad study and though at first it was the topic given to me, I’ve grown to deeply love the subject which has been the centre of my life this past one year and will be for the coming two years! And yeah as I said now I am in love with this research so I want to do something worthy enough for people suffering from this disease.

I deal with samples obtained from CML patients who come to the hospital for a regular check up and are on IM. Informed consent is obtained from each n every patient for this research. Patients who do not respond to IM have been administered alternative treatment but at this stage the treatment regime is not exactly the core of my concern but rather, the cause of “failure” of the treatment! I hope u hold a solution for my sequencing problems. I have tried to follow all the protocols published to the letter but have been unsuccessful thus far. Thanks for your support.

Please hold for a few days! I am arranging help! My Guide is a nervous and unsecured guy; scientists as you know are usually unsecured due to difficulty in arranging for funding. I will talk to him n request to help you! By the way, can your team agree for any kind of collaboration with us? That way he will help you whole heartedly! Don’t lose heart! Hope you will stay in Leukemia research; we need more and more people!
